Potentiality of statistical experimental design in optimization of effective parameters on extraction of essential oil from Rosa damascene Mill

چکیده مقالهThe essential oil content of the oil-bearing rose (Rosa damascene Mill ) is relatively low, around 0.3-0.4 mLkg-1 in fresh flowers. Therefore, there is a high interest to find modified methods to increase extraction yield of essential oil of the mentioned plant [1]. In the present study, the effect of storage conditions of flowers on content and composition of the Rosa damascene essential oil was investigated to find the optimum conditions to save flowers for longer time without significant change in quality and quantity of the oil. The rose oil was obtained by hydro-distillation in Clevenger type apparatus and the components in the oil were analyzed by GC-MS. Eleven experiments with two influencing parameters (temperature and time of storage) with five levels were performed based on central composite design (CCD) and response surface methodology (RSM) to evaluate the individual and interactive effects of the effective variables [2, 3]. CCD was employed for the experimental design and RSM was applied for building and analyzing the model. The structure of central composite design allows the development of a second-order polynomial model with regard to the set of linear, interaction and quadratic effects. Based on the obtained results from analysis of variance (ANOVA) and response surface plot in order to evaluate the suitability and adequacy of a quadratic model, the changes in the mentioned parameters have high effects on both quantity and quality of the essential oil. Therefore, temperature (between -5 and 15 oC) and time of storage (between 1 and 14 days) as effective parameters were optimized in order to obtain the highest quantity and the best quality of the essential oil.
