Abstract

Heparin, a sulfated glycosaminoglycan, is widely used as an anticoagulant in clinical medicine. However, conventional production from animal tissues presents limitations including microbial and viral contamination risks, structural variability, and scalability constraints. In recent years, advances in biotechnology—such as microbial and mammalian cell systems, engineered enzymatic pathways, and biocatalysts—have enabled the production of recombinant heparin with controlled molecular weight and sulfation patterns. Beyond its classical anticoagulant function, heparin demonstrates antiviral and anti-inflammatory activities and shows potential in regenerative medicine and nanocarrier-based drug delivery. Despite significant progress, challenges remain regarding production costs, precise molecular structure control, scalability, and regulatory hurdles. Future directions emphasize enzymatic pathway engineering, nanobiotechnology integration, and the development of targeted delivery systems, aiming to enhance the efficacy and safety of heparin therapy. This review highlights recent advancements, therapeutic applications, current limitations, and future perspectives in the biotechnological production of novel heparin.
