چکیده مقاله
Bacteria can break down this organic material and obtain energy and building material for cell growth. Wastewater treatment plants try to optimize this ability of bacteria to eat the organics. This research was conducted with the purpose of studying the pollution of sludge derived from wastewater rosewater factory which can be used of microorganisms for wastewater treatment. In this study, samples were collected in an aseptic manner. Serial dilutions of the samples up to 10-6 were prepared for proper isolation of individual colony forming units. Bacterial isolates were cultured on LB medium and incubation at 30oC for 24 hours. Phenotypic characteristics of individual isolates including Gram-staining, cell morphology, catalase and oxidase activities were determined using standard methods. After several dilutions and repeated subculturing, 75 pure bacterial cultures isolated including 7 gram-positive bacilli, 12 gram-negative bacilli, 5 gram-positive cocci , 14 gramnegative cocci , 6 gram-positive coccobacilli , 4 bacteria oval , 21 gram- negative coccobacilli Polymorph , 5 fungi and one yeast. Morphological variation was observed when colonies were compared including smooth, circular, low-convex, transparent or translucent and different colors like red, orange-red, pale-pink, yellowish, cream or white. Sixty five isolates were catalasepositive, 4 catalase-negative, 10 oxidase-positive and 59 oxidase-negative. The microorganisms-associated habitat is a dynamic environment in which many factors may affect the structure and species composition of the bacterial communities that colonize in the wastewater plants.
