Analysis of the expression of heat shock protein Hsp۹۰ in response to polymeric nano-cucrcumin on gastric cancer cell line (AGS)

چکیده مقاله

Introduction: The expression levels of heat shock proteins (Hsps) areimpaired in many types of cancers. This impairment is of ten associated with poor prognosis for sur vival and response to treatment. It is also accompanied by cell proliferation, invasion and metastasis. Curcuminhas anti - carcinogenic, anti - proliferation, anti - inflammatory , anti - mutagenic, anti - angiogenesis andantioxidant properties. Curcumin has also ant i - invasive properties through, at least in part, regulation of heat shock protein genes. Methods: Gastric cancer cell line (AGS) was treated with effective dose of polymeric nano - curcumin in triplicate, and PBS treatment in parallel as control. The effect ive does of drug was determined by MTT assay, as a concentration of drug that causes 50% cell growth inhibition (IC 50 ). The expression levels of hsp90 mRNA then assessed by semi - quantitative RT - PCR, using ...........gene as reference. Results: Comparing the band sharpness of RT - PCR products from treated and non - treated flask, it is obvious that the expression levels of Hsp90 mRNA were significantly elevated by polymeric nano - curcumin treatment. The data of replicas are in concordance. Conclusion: Overall, our data showed that polymeric nano - curcumin could disturb the HSP content of cell, especially the HSP90. This disturbance could be deciphered as a consequence of cellular resistance against toxic effects of polymeric nano - curcumin, or participating in apoptosis induction by this drug.
