Investigation of serum cholestrol level in the individuals affected by obesity and correlation of it with rs2241766 and rs266729 polymorphisms of adipoq gene

Background: Undoubtedly, obesity is one of the most common diseases in the community, with its side effects, such as metabolic aberration, liver and heart disease. So far, several studies have been conducted on the association of ADIPOQ gene polymorphisms with obesity and hypo-adiponctinemia in different population. Adiponectin is a protein hormone involved in regulating of fatty acid breakdown that is encoded by the ADIPOQ gene. Material and methods: In this study, blood samples were taken from the individuals registered to the TAMIN EJTEMAEE laboratory of Borujen city who were referred with obesity problems. LDL and free cholesterol level was analyzed on the blood of the patients using the El200 auto-analyzer. In the next step, Total DNA was extracted from the blood samples and subjected to RFLP and tetra-ARMS methods in order to investigation of rs2241766 and rs266729 polymorphisms of ADIPOQ gene respectively. Result and discussion: Our results confirmed the association of these polymorphisms at a significant level (p <0.05). The rs266729 polymorphism is located in the promoter region of ADIPOQ Gene. The presence of the G allele is likely to reduce the tendency of the transcription factors at -11377 position, which in turn can reduce the level of ADIPOQ expression and the related effects. Also, rs2241766 polymorphism showed a significant correlation with elevated cholesterol levels in the presence of T allele in this position from the exon 2 of ADIPOQ gene encoding adiponectin. In this case, a significant reduction in codon preference can reduce the expression of adiponectin protein. Our result can introduce two SNP marker applicable to the screening for high risk individuals for hypercholesterolemia.
