The Phylogeny of the genus Moltkia Lehm. (Boraginaceae) from Iran based on plastid trnL-trnF region sequence data

چکیده مقاله

The angiosperm family Boraginaceae includes ca. 1600 species distributed among ca. 110 genera. Moltkia Lehm. (Boraginaceae tribe Lithospermeae) comprises about nine species. Our study applies molecular phylogenetic methods to test the monophyly and relationships of Moltkia. We used DNA sequence data from the Plastid trnL-trnF region for study. Ten specimens from 4 species of Moltkia were collected from their natural habitats in Iran. Total genomic DNA was isolated from dried leaves using the standard CTAB extraction protocol. The trnL-trnF region of each sample were amplified by PCR employing trnLUAAF (TabC) and trnFUAAR (TabF) primers. Anchusa formosa Selvi & al. and Anchusa crispa Viv. were included in the analyses as outgroups. Sequences were aligned with Bioedit Vers. 7.0.9. and adjusted manually. The aligned data matrix was reconstructed by parsimony optimality criterion using PAUP* ver. 4.0b10. Support for clades were evaluated by bootstrapping using 1000 replications with the heuristic search option. The aligned data matrix (with outgroup) consisted of 972 characters, of which 916 characters are constant; 3 variable characters are parsimony-uninformative and 53 parsimony- informative characters were detected. The monophyly of Moltkia is corroborated by high bootstrap support value of 100 %. From 872 total characters of ingroup, 865 characters are constant. The ingroup falls into two main clades. All studied species except one population constitute one main clade with BS of 73% and one population of M. coerulea separated from other population with BS under 50%.
